Additionally, M2 macrophages can be subdivided into M2a, M2b, M2c, and M2d phenotypes, linked to different inducers but sharing similar functional activities. Functionally, macrophages can be polarized into two extreme groups: classical M1 with pro-inflammatory and anti-tumorigenic functions and alternative M2, which act as anti-inflammatory and pro-tumorigenic. Tumor associated macrophages (TAMs) are the most abundant immune cells of the TME. As for the tumor cells, genetic alterations resulting in uncontrolled proliferation and phenotype changes are characteristic, the stromal cells remain genetically unchanged, but their activity is largely influenced by cytokines produced by tumor cells themselves or by other cells of the TME.
